چکیده

The effects of gaseous anesthetics and pressure on the thermotropic behavior of multilamellar dipalmitoyl-phosphatidylcholine liposomes were studied by using a high-sensitivity differential scanning calorimeter. It was found that halothane and enflurane decreased the transition temperature and increased the width of the transition without affecting the enthalpy change for the main gel-to-liquid crystalline transition. This demonstrated that the anesthetics decreased the degree of cooperative interaction between phospholipid molecules within the bilayer. Increasing the pressure increased the transition temperature but did not affect the transition width or enthalpy change. However, the increase in pressure reversed the effect of anesthetic on both the transition temperature and transition width. It is suggested that an understanding of the effect of anesthetics on the degree of cooperative interaction between phospholipids may be a key to understanding anesthetic action.

A scanning calorimetric study of the interaction of anthracyclines with neutral and acidic phospholipids alone and in binary mixtures.

High sensitivity differential scanning calorimetry was employed to study the thermotropic behavior of multilamellar vesicles of neutral and acidic phospholipids and binary mixtures thereof in the presence of anthracycline antibiotics. Adriamycin and its lipophilic analogue, N-trifluoroacetyladriamycin-14-valerate (AD32) were investigated and compared to chlorpromazine and quinidine with respect...

Formation and Physicochemical Properties of Liposomes Using a Supercritical Reverse Phase Evaporation Method

L-a-dipalmitoylphosphatidylcholine (DPPC) liposomes were prepared using supercritical reverse phase evaporation (scRPE) method. The effects of phospholipid structure, preparation pressure, and amount of ethanol on their physicochemical properties were examined using measurements of trapping efficiency, cloud point measurements, dynamic light scattering, and osmotic water transport studies. The ...

Effect of sodium cholate on the phase transition temperature of dipalmitoyl phosphatidylcholine.

On treatment with sodium cholate, the phase transition temperature (Tc) of multilamellar liposomes derived from dipalmitoyl phosphatidylcholine (DPPC) progressively decreased with increasing cholate: lipid-P molar ratio. A molar ratio of 4.5 caused a one degree depression in Tc. The unilamellar cholate vesicles of DPPC exhibited the same Tc as that of the multilamellar ones. The formation of un...

Cooperative effects in the interaction between melittin and phosphatidylcholine model membranes. Studies by temperature scanning densitometry.

The interaction between the peptide melittin from bee venom with multilamellar liposomes of dipalmitoylphosphatidylcholine or egg yolk phosphatidylcholine has been studied by the method of temperature scanning densitometry, yielding information on the specific volume of the association products and on the changes during the thermotropic transition of the lipids. The effects of the interaction w...

Local anesthetics induce interdigitation and thermotropic changes in dipalmitoylphosphatidylcholine bilayers.

The molecular mechanism underlying the action of local anesthetics is still elusive. Phenylethanol (PEtOH) is an ingredient of essential oils with a rose-like odor and has been used as a local anesthetic. In this work, we have explored the effect of PEtOH on thermotropic behavior and organization of dipalmitoylphosphatidylcholine (DPPC) membranes utilizing differential scanning calorimetry (DSC...
